SELP Gene

HGNC Family CD molecules (CD)
Name selectin P (granule membrane protein 140kDa, antigen CD62)
Description This gene encodes a 140 kDa protein that is stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells. This protein redistributes to the plasma membrane during platelet activation and degranulation and mediates the interaction of activated endothelial cells or platelets with leukocytes. The membrane protein is a calcium-dependent receptor that binds to sialylated forms of Lewis blood group carbohydrate antigens on neutrophils and monocytes. Alternative splice variants may occur but are not well documented. [provided by RefSeq, Jul 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nSELP, the gene encoding P‑selectin, is a critical adhesion receptor expressed on activated endothelial cells and platelets. Upon stimulation, P‑selectin is externalized from Weibel–Palade bodies and α‑granules, where it mediates the very first steps of leukocyte recruitment in inflamed vascular beds by forming reversible bonds with specialized glycoprotein ligands—such as PSGL‑1, CD44, and ESL‑1—that promote leukocyte tethering and rolling. These interactions also trigger intracellular signals that contribute to the “slow rolling” state necessary for integrin activation and firm adhesion, thereby coordinating the inflammatory response."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nApart from its role in leukocyte trafficking, SELP is intimately involved in thrombus formation. P‑selectin contributes to the generation of procoagulant microparticles via engagement with its ligand PSGL‑1, a process that not only localizes tissue factor but also accelerates fibrin formation at sites of vascular injury. Its ability to both mediate platelet–endothelial as well as platelet–leukocyte interactions underscores its dual role in linking inflammation with thrombosis, as evidenced by experimental studies that demonstrate improved hemostasis in bleeding disorders with controlled P‑selectin activity."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCirculating levels of soluble P‑selectin (sP‑selectin) not only serve as a marker of vascular activation but also have predictive value for thrombotic complications in various clinical settings. Elevated sP‑selectin levels have been linked to an increased risk of venous thromboembolism in cancer patients, and genome‐wide as well as candidate gene association studies have identified polymorphisms within SELP that modulate its plasma concentration and contribute to cardiovascular risk. These findings highlight the potential of sP‑selectin as a biomarker to stratify patients according to thrombotic risk."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its hemostatic and inflammatory roles, SELP is increasingly recognized for its contribution to pathological processes such as cancer metastasis and complement‐dependent microvascular thrombosis. For example, P‑selectin is implicated in the adhesion of activated platelets to tumor cells, which facilitates tumor cell survival in the circulation and subsequent metastatic seeding. Moreover, up‐regulation of endothelial P‑selectin by bacterial toxins or inflammatory mediators can activate the complement cascade and exacerbate local thrombosis in conditions like hemolytic uremic syndrome, underscoring the molecule’s multifaceted role in disease."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "12", "end_ref": "16"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nRecent mechanistic and genetic studies have expanded our understanding of the regulation of SELP and its interactions with the actin cytoskeleton, as well as with other key molecules such as von Willebrand factor. Animal models and transgenic approaches have demonstrated that both the cellular and soluble forms of P‑selectin are amenable to modulation by pharmacologic agents like hydroxyurea, which can mitigate its prothrombotic effects. Furthermore, novel insights into the targeting of intracellular trafficking pathways emphasize the therapeutic potential of disrupting SELP-mediated adhesion in diverse conditions ranging from inflammatory diseases to atherothrombosis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "17", "end_ref": "25"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Alexander Zarbock, Klaus Ley, Rodger P McEver, et al.
