Description The reversible posttranslational modification of proteins by the addition of small ubiquitin-like SUMO proteins (see SUMO1; MIM 601912) is required for numerous biologic processes. SUMO-specific proteases, such as SENP5, are responsible for the initial processing of SUMO precursors to generate a C-terminal diglycine motif required for the conjugation reaction. They also have isopeptidase activity for the removal of SUMO from high molecular mass SUMO conjugates (Di Bacco et al., 2006 [PubMed 16738315]).[supplied by OMIM, Jun 2009]

In oral squamous cell carcinoma, its subcellular localization shifts from the nucleolus to the cytoplasm and correlates with the degree of tumor differentiation. In breast cancer, reduced SENP5 expression is associated with a better prognosis, while its silencing diminishes malignant traits such as anchorage-independent growth, proliferation, migration, and invasion via regulation of the TGFβ receptor I–MMP9 cascade. In oral squamous cell carcinoma cells, SENP5 stabilization under mild oxidative stress protects against apoptosis by maintaining mitochondrial integrity. In a developmental setting, gestational zinc deficiency leads to SENP5 overexpression, which correlates with reduced SUMO1 conjugation, abnormal cardiomyocyte differentiation, increased apoptosis, and ultimately cardiac dysplasia.
