SGCE Gene

Name sarcoglycan, epsilon
Description This gene encodes the epsilon member of the sarcoglycan family. Sarcoglycans are transmembrane proteins that are components of the dystrophin-glycoprotein complex, which link the actin cytoskeleton to the extracellular matrix. Unlike other family members which are predominantly expressed in striated muscle, the epsilon sarcoglycan is more broadly expressed. Mutations in this gene are associated with myoclonus-dystonia syndrome. This gene is imprinted, with preferential expression from the paternal allele. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. A pseudogene associated with this gene is located on chromosome 2. [provided by RefSeq, Oct 2016]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nEpsilon‐sarcoglycan, encoded by SGCE, is a transmembrane protein that contributes to the integrity and function of neuronal membranes. Its expression is ubiquitous throughout the brain—including the basal ganglia and cerebellum—and its regulation is unusual because the SGCE gene is subject to maternal imprinting via differential methylation. In consequence, only the paternally‐derived allele is expressed in key brain regions, a feature that has been documented in several studies and underpins the autosomal‐dominant inheritance pattern with reduced penetrance seen in myoclonus‐dystonia syndrome."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "15"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAt the cellular level, proper trafficking of epsilon‐sarcoglycan to the plasma membrane is crucial for its function. Disease‐associated SGCE mutations are frequently loss‐of‐function variants that impair this trafficking; mutant proteins are misfolded, sequestered intracellularly, become polyubiquitinated, and are rapidly degraded by the proteasome. Such defects have been demonstrated by studies assessing altered biosynthesis and subcellular localization, and they suggest that disruption of membrane targeting and protein stability is central to the pathogenesis of myoclonus‐dystonia."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "16", "end_ref": "30"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nThe disruption of SGCE function due to various mutations (including point mutations, exon deletions, and copy‐number variations) leads to the distinct phenotype of myoclonus‐dystonia, frequently accompanied by psychiatric disturbances such as obsessive–compulsive symptoms and anxiety. Moreover, differential expression of brain‐specific SGCE isoforms—most notably in cerebellar Purkinje cells and dentate nucleus neurons—supports a model in which dysfunction of specific neural circuits contributes to the observed motor and non‐motor manifestations.
