| HGNC Family | F-boxes (FBX) |
| Name | S-phase kinase-associated protein 2, E3 ubiquitin protein ligase |
| Description | This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbls class; in addition to an F-box, this protein contains 10 tandem leucine-rich repeats. This protein is an essential element of the cyclin A-CDK2 S-phase kinase. It specifically recognizes phosphorylated cyclin-dependent kinase inhibitor 1B (CDKN1B, also referred to as p27 or KIP1) predominantly in S phase and interacts with S-phase kinase-associated protein 1 (SKP1 or p19). In addition, this gene is established as a protooncogene causally involved in the pathogenesis of lymphomas. Alternative splicing of this gene generates three transcript variants encoding different isoforms. [provided by RefSeq, Jul 2011] |

Early structural studies have revealed that SKP2, in complex with Skp1 and Cul1, provides the substrate–recognition module necessary for targeting a variety of cell cycle regulators for ubiquitin‐dependent degradation. In particular, SKP2 directs the turnover of inhibitors of cyclin–dependent kinases—such as p27Kip1, p57Kip2, and the pocket protein p130—which in turn facilitates the G1–S phase transition and proper DNA recombination processes, as exemplified by RAG-2 degradation."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nUpstream, SKP2 expression and activity are tightly regulated by various signaling pathways and post‐translational modifications. Cell shape and RhoA signaling can promote SKP2 expression, while oncogenic transcription factors—such as MYC—directly induce SKP2 transcription. Furthermore, SKP2 abundance and activity are controlled by modifications including neddylation, phosphorylation and acetylation. For instance, Akt-dependent phosphorylation events help prevent its degradation via APC/C(Cdh1), whereas acetylation by p300 enhances its stability and promotes cytoplasmic localization linked to increased cell migration. In addition, tumor suppressors such as FOXP3 and signaling effectors including IKKα counteract SKP2 expression, and p53 pathways are antagonized by SKP2 to suppress apoptosis. Collectively, these regulatory inputs—from cell shape cues to kinase cascades and transcriptional repressors—finely tune SKP2’s function during cell cycle progression and determine its oncogenic potential."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "19"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nDownstream, SKP2 exerts potent oncogenic functions by targeting multiple substrates that modulate proliferation, survival, and DNA repair. Beyond degrading cell cycle inhibitors, SKP2 can regulate signaling pathways by facilitating ubiquitination of key proteins such as Akt and FOXO transcription factors. It also modulates autophagy by catalyzing K48-linked ubiquitination of Beclin1, and influences the DNA damage response through K63-linked ubiquitination of NBS1, thereby promoting ATM activation. Additional studies have implicated SKP2 in controlling the activity of oncoproteins like YAP and in the polyubiquitination of LKB1, mechanisms that further link its activity to tumor progression and metabolic stress responses.
