Description SLC2A12 belongs to a family of transporters that catalyze the uptake of sugars through facilitated diffusion (Rogers et al., 2002). In skeletal muscle, for example, its substantial mRNA expression—especially in type I oxidative fibers—implies a contribution to sustaining aerobic metabolism. In the kidney, GLUT12 is localized predominantly to the distal tubules and collecting ducts with an apical distribution that suggests a role in augmenting late-nephron glucose reabsorption. In adipose tissues, GLUT12 is present in mature adipocytes and its expression is regulated by hormonal stimuli; similar observations have been made in studies of 3T3-L1 adipocytes as well as adipose depots from lean and diet-induced obese mice. Furthermore, in the small intestine, GLUT12 translocates from the apical cytoplasmic pool to the brush border membrane in response to glucose and insulin, thereby contributing to postprandial sugar absorption. In triple-negative breast cancer, for instance, upregulation of GLUT12 promotes tumor growth and metastasis by enhancing aerobic glycolysis, while in gastric cancer an androgen receptor–dependent increase in GLUT12 expression counteracts the anticancer effects of targeted therapy. The transcription factor TWIST1 has also been shown to increase GLUT12 expression, facilitating insulin-independent glucose uptake that may support the metabolic demands of cancer cells.
