SPN Gene

HGNC Family CD molecules (CD)
Name sialophorin
Description This gene encodes a highly sialylated glycoprotein that functions in antigen-specific activation of T cells, and is found on the surface of thymocytes, T lymphocytes, monocytes, granulocytes, and some B lymphocytes. It contains a mucin-like extracellular domain, a transmembrane region and a carboxy-terminal intracellular region. The extracellular domain has a high proportion of serine and threonine residues, allowing extensive O-glycosylation, and has one potential N-glycosylation site, while the carboxy-terminal region has potential phosphorylation sites that may mediate transduction of activation signals. Different glycoforms of this protein have been described. In stimulated immune cells, proteolytic cleavage of the extracellular domain occurs in some cell types, releasing a soluble extracellular fragment. Defects in expression of this gene are associated with Wiskott-Aldrich syndrome. [provided by RefSeq, Sep 2017]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nSPN (also known as CD43 or leukosialin) is an abundant transmembrane sialoglycoprotein that plays multifaceted roles in hematopoietic differentiation and immune cell function. In early hematopoietic development, CD43 serves as a critical marker that distinguishes pre‐committed progenitors from endothelial or mesenchymal cells, reflecting its importance in lineage commitment. Equally, in mature leukocytes its dynamic cell surface expression helps regulate intercellular adhesion, costimulatory signals, apoptosis, and the fine‐tuning of T‐cell receptor–mediated activation through modulation of kinase phosphorylation and adapter recruitment. These integrated signaling processes ultimately influence cytokine production and cell proliferation, ensuring that immune responses are both robust and tightly controlled."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCD43 also has a central role in host–pathogen interactions and the regulation of inflammation. Bacterial virulence factors—notably the class‐2 SPATE proteases—target CD43 (along with other adhesion receptors) to modulate leukocyte trafficking and dampen chemotaxis. In viral infections, such as those involving HTLV-1 and HIV-1, CD43 is involved in the assembly and incorporation of viral particles as well as the modulation of cell–cell transmission. In addition, proteolytic shedding of CD43 from activated neutrophils by enzymes like cathepsin G alters its extracellular domain, thereby affecting interactions with endothelial molecules and inhibitory receptors (for example, Siglec-7), which in turn regulates immune synapse organization and cytotoxic function."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "16"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the context of cancer biology, aberrant expression and glycosylation of CD43 have emerged as key diagnostic and prognostic markers in both hematopoietic and nonhematopoietic malignancies. Altered CD43 expression has been linked to the modulation of cell adhesion, migration, and survival; for instance, its interplay with tumor suppressor pathways (including p53 and ARF) influences apoptotic responses and proliferative control. Moreover, unique CD43 glycoforms (such as those recognized by the UN1 antibody) have been exploited as immunophenotypic targets in lymphoma, colon carcinoma, and other solid tumors, aiding in both differential diagnosis and in stratification of clinical prognosis.
