| HGNC Family | Serine/arginine-rich splicing factors (SRSF), RNA binding motif containing (RBM) |
| Name | serine/arginine-rich splicing factor 9 |
| Description | The protein encoded by this gene is a member of the serine/arginine (SR)-rich family of pre-mRNA splicing factors, which constitute part of the spliceosome. Each of these factors contains an RNA recognition motif (RRM) for binding RNA and an RS domain for binding other proteins. The RS domain is rich in serine and arginine residues and facilitates interaction between different SR splicing factors. In addition to being critical for mRNA splicing, the SR proteins have also been shown to be involved in mRNA export from the nucleus and in translation. Two pseudogenes, one on chromosome 15 and the other on chromosome 21, have been found for this gene. [provided by RefSeq, Sep 2010] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nSRSF9, also known as SRp30c, is a multifunctional splicing factor that regulates alternative pre‐mRNA splicing by binding specific cis‐elements within target transcripts. Multiple studies have shown that SRSF9 directly modulates the splicing of key mediators of apoptosis and stress response. For example, its binding to defined intronic elements controls the splicing choices in genes such as Bcl‑x and Caspase‑2, thereby influencing the production of isoforms with opposite functions in programmed cell death. In addition, SRSF9 is required for the alternative splicing of glucocorticoid receptor pre‑mRNA, favoring generation of the GRβ isoform that can act as a dominant negative regulator—an effect exploited in conditions such as glucocorticoid resistance seen in inflammatory and cancer cells. Furthermore, SRSF9 interacts with transcripts like those of hnRNP A1 to modulate exon skipping events and is subject to regulation by tumor suppressive microRNAs (e.g., miR‑1) which, when expressed, reduce SRSF9 levels and promote apoptosis in cancer models."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its critical role in splicing regulation, aberrant SRSF9 expression has been closely linked with tumorigenesis. Overexpression of SRSF9 promotes oncogenic signaling by enhancing the translation of β‑catenin mRNA in an mTOR‑dependent manner, thereby contributing to its accumulation in cancer cells. This splicing factor is frequently upregulated in a range of malignancies—including colorectal cancer, hepatocellular carcinoma and glioblastoma—and its modulation by microRNAs (for instance, miR‑802) has been shown to attenuate cancer cell proliferation and invasion. Moreover, in certain viral models such as human papillomavirus (HPV‑16) and HIV‑1, SRSF9 can redirect splicing patterns to favor production of viral mRNAs that are critical for late gene expression, highlighting its versatile regulatory capacities in both host and viral gene expression programs."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nSRSF9 function is further influenced by post‑translational modifications and contextual cellular cues. Its subnuclear localization is determined by arginine methylation, while phosphorylation—such as by AMPK—modulates its affinity for target mRNAs like that of β‑catenin, thereby fine‑tuning protein synthesis. In addition, SRSF9 has been shown to repress adenosine‑to‑inosine RNA editing by interacting with ADAR2, and it plays a corrective role in aberrant splicing events witnessed in monogenic disorders such as aromatic l‑amino acid decarboxylase deficiency. Notably, formation of nuclear stress bodies during thermal stress also recruits kinases that re‑phosphorylate SRSF9, underscoring its dynamic role in the cellular stress response."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "19", "end_ref": "22"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Martin J Simard, Benoit Chabot "}, {"type": "b", "children": [{"type": "t", "text": "SRp30c is a repressor of 3' splice site utilization."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Mol Cell Biol (2002)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1128/MCB.22.12.4001-4010.2002"}], "href": "https://doi.org/10.1128/MCB.22.12.4001-4010.2002"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "12024014"}], "href": "https://pubmed.ncbi.nlm.nih.gov/12024014"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Ute Raffetseder, Björn Frye, Thomas Rauen, et al.
