| HGNC Family | G protein-coupled receptors |
| Name | somatostatin receptor 5 |
| Description | Somatostatin and its related peptide cortistatin exert multiple biological actions on normal and tumoral tissue targets by interacting with somatostatin receptors (SSTRs). The protein encoded by this gene is one of the SSTRs, which is a multi-pass membrane protein and belongs to the G-protein coupled receptor 1 family. The activity of this receptor is mediated by G proteins which inhibit adenylyl cyclase, and different regions of this receptor molecule are required for the activation of different signaling pathways. A mutation in this gene results in somatostatin analog resistance. Alternatively spliced transcript variants have been identified in this gene.[provided by RefSeq, Feb 2010] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nSomatostatin receptor subtype 5 (SSTR5) is widely expressed in a spectrum of endocrine and neuroendocrine tumors, where its distribution has been linked to the regulation of hormone secretion and tumor growth. In several studies, high SSTR5 expression was demonstrated in somatostatinomas, pituitary adenomas (including growth hormone‐ and TSH‐secreting tumors), insulinomas, and other gastroenteropancreatic lesions – findings that have been instrumental in guiding both diagnostic imaging and the use of somatostatin analogs to control hormonal hypersecretion and tumor progression. Differential receptor expression patterns among tumor subtypes have further suggested that the balance of SSTR5 with other SSTR subtypes may help predict the effectiveness of therapeutic agents and inform tailored treatment strategies in conditions such as acromegaly and Cushing’s disease."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "24"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAt the molecular level, SSTR5 operates as a seven‐transmembrane G protein–coupled receptor whose function is intricately regulated by receptor–receptor interactions, post‐translational modifications, and precise intracellular signaling events. SSTR5 can form both homodimers and heterodimers (for example, with SSTR1, SSTR2, or even β₁‐adrenergic receptors), an interplay that modulates its trafficking, receptor internalization, and recycling. Specific intracellular domains—including a PDZ ligand motif that interacts with scaffolding proteins, a conserved DRY motif, and a BBXXB region in the third intracellular loop—are critical for coupling to downstream effectors. Moreover, agonist–induced phosphorylation (mediated via G protein–coupled receptor kinases and subsequently reversed by protein phosphatases) and reversible palmitoylation (by enzymes such as ZDHHC5) fine–tune SSTR5’s capacity to inhibit adenylyl cyclase activity and modulate MAPK and PI3K/AKT pathways."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "25", "end_ref": "35"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditional layers of complexity in SSTR5 biology arise from the emergence of truncated receptor variants and genetic polymorphisms that can markedly alter receptor function and influence tumor behavior. Novel splice variants—such as sst5TMD4—have been identified in various tumor types including pituitary, thyroid, prostate, and breast cancers; these aberrant forms not only disrupt normal somatostatin signaling but are also associated with enhanced cell proliferation, increased invasiveness, and resistance to conventional somatostatin analog therapies. In parallel, polymorphic variants in the SSTR5 gene have been linked to differential hormone levels and may underlie variable responses to treatment, underscoring the potential utility of SSTR5 as both a prognostic biomarker and a therapeutic target in a range of endocrine malignancies."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "36", "end_ref": "42"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "M Papotti, M Bongiovanni, M Volante, et al.
