In one study, STK11IP was shown to participate in a protein complex with LKB1 and Smad4, wherein LKB1 phosphorylates Smad4 on a critical threonine residue. This phosphorylation impairs Smad4’s ability to bind to promoter elements of TGFβ and bone morphogenetic protein target genes, thereby negatively regulating Smad4‐dependent transcription and subsequent epithelial–mesenchymal transition. This mechanism highlights a role for STK11IP in fine‐tuning TGFβ signaling responses in the cell."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "In a separate investigation of cardiac pathology, STK11IP was found to be significantly up‐regulated in both human heart failure samples and mouse models of hypertrophy. Here, STK11IP contributes to pathological cardiac hypertrophy by directly targeting the lipid phosphatase PTEN, thereby inhibiting its activity and promoting Akt signaling. In this context, enhanced Akt pathway activation drives not only cardiomyocyte hypertrophy but also fibrosis, whereas STK11IP ablation markedly protects against these deleterious structural and functional changes.
