TIRAP Gene

Name toll-interleukin 1 receptor (TIR) domain containing adaptor protein
Description The innate immune system recognizes microbial pathogens through Toll-like receptors (TLRs), which identify pathogen-associated molecular patterns. Different TLRs recognize different pathogen-associated molecular patterns and all TLRs have a Toll-interleukin 1 receptor (TIR) domain, which is responsible for signal transduction. The protein encoded by this gene is a TIR adaptor protein involved in the TLR4 signaling pathway of the immune system. It activates NF-kappa-B, MAPK1, MAPK3 and JNK, which then results in cytokine secretion and the inflammatory response. Alternative splicing of this gene results in several transcript variants; however, not all variants have been fully described. [provided by RefSeq, Jul 2008]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nTIRAP (TIR‐domain–containing adaptor protein), also known as MAL, is a pivotal adaptor in innate immune signaling that links activated Toll‐like receptors (TLR2 and TLR4) to downstream effectors. It is recruited to the plasma membrane by virtue of a phosphatidylinositol 4,5‐bisphosphate (PIP₂)–binding domain, an event that facilitates the delivery of MyD88 to engaged TLRs and the initiation of signaling cascades leading to NF‑κB and MAP kinase activation. In this context, TIRAP orchestrates early events in the innate immune response by determining the specificity and amplitude of the signal and by acting as a critical “bridge” between the receptor and intracellular signaling complexes."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its membrane recruitment, TIRAP’s TIR domain engages in a variety of protein–protein interactions that are essential for effective signaling. Structural and mutational studies have demonstrated that TIRAP possesses unique features—such as an unusually long AB loop, the capacity to form dimers or even reversible filaments, and specific motifs that enable binding to partners like MyD88 and TRAF6. Post‐translational modifications including tyrosine phosphorylation (for example, via Bruton's tyrosine kinase) further regulate these interactions, fine‐tuning NF‑κB transactivation and other downstream responses. Certain mutations (e.g., D96N) disrupt the ability of TIRAP to interact with MyD88, underscoring the precise molecular requirements for its adaptor function, while its participation extends to crosstalk with additional receptors such as RAGE."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "20"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nClinically, genetic variations in TIRAP have been associated with differential susceptibility to a variety of infectious and inflammatory diseases—including tuberculosis, sepsis, Chagas cardiomyopathy, autoimmune disorders, and altered responses in conditions such as atopic dermatitis and rheumatoid arthritis.
