TNFRSF4 Gene

HGNC Family CD molecules (CD), Tumor necrosis factor receptor superfamily (TNFRSF)
Name tumor necrosis factor receptor superfamily, member 4
Description The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor has been shown to activate NF-kappaB through its interaction with adaptor proteins TRAF2 and TRAF5. Knockout studies in mice suggested that this receptor promotes the expression of apoptosis inhibitors BCL2 and BCL2lL1/BCL2-XL, and thus suppresses apoptosis. The knockout studies also suggested the roles of this receptor in CD4+ T cell response, as well as in T cell-dependent B cell proliferation and differentiation. [provided by RefSeq, Jul 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nTNFRSF4 (also known as OX40 or CD134) plays a central costimulatory role during T‐cell activation by augmenting proliferation, survival, and effector differentiation. Engagement of TNFRSF4 by its ligand facilitates key intracellular signaling cascades—including recruitment of PI3K and PKB—that boost clonal expansion and promote the differentiation and persistence of both CD4+ helper and CD8+ memory T cells. These signals not only enhance antigen‐driven cytokine production but also support long‐term immune memory and resistance to apoptosis, as evidenced by findings in models of infection, autoimmunity, and cancer immunotherapy."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its positive role in amplifying conventional T‐cell responses, TNFRSF4 signaling influences immune regulation in diverse pathological settings. Its activity modulates the balance between effector and regulatory T cells in conditions such as systemic lupus erythematosus, allergic airway inflammation, and various autoimmune disorders, where its expression correlates with disease activity and tissue inflammation. Furthermore, TNFRSF4 engagement can alter the phenotype of regulatory T cells and shape the cytokine milieu in chronic inflammatory diseases, a feature that has also been linked to cardiovascular complications and metabolic disturbances seen in obesity and type 2 diabetes."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "12", "end_ref": "26"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its immunomodulatory influence on T‐cell function, TNFRSF4 also serves as a viral entry receptor and a promising therapeutic target in cancer and immunotherapy. In certain viral infections—such as human herpesvirus 6B (HHV‑6B)—CD134 functions as a critical entry receptor through defined cysteine‐rich domains, while its ligand interactions and oligomerization status significantly influence downstream signaling. In parallel, agonistic antibodies targeting TNFRSF4 are under clinical investigation to harness its costimulatory capacity for enhancing antitumor immunity, with studies demonstrating its ability to modulate natural killer and dendritic cell functions and potentiate tumor‐specific T‐cell responses."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "27", "end_ref": "34"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Susanna Carboni, Fahmy Aboul-Enein, Caroline Waltzinger, et al.
