Mechanistically, the ZNF689-TRIM28 complex normally binds to the promoter of LINE-1 elements, inducing H3K9me3-mediated transcriptional silencing to prevent retrotransposition and maintain genomic stability. Loss of ZNF689 reactivates LINE-1 activity, exacerbating genomic instability and fostering heterogeneous tumor cell populations that impair antigen presentation and T-cell activation.
